The protein conversation in performance nutrition has improved but remains anchored on total daily intake. Most guidance says 1.6-2.2g per kilogram of bodyweight. That range is broadly correct. It is also incomplete.
How you distribute protein across the day, and the quality of each serving, determines how much of your total intake actually drives muscle protein synthesis.
The Distribution Problem
Muscle protein synthesis is not a continuous process proportional to total intake. It is a pulsatile response driven by leucine availability at each meal. Research consistently shows that multiple moderate protein doses throughout the day produce greater total synthesis than the same total protein in fewer, larger servings.
The optimal dose per meal for maximal muscle protein synthesis stimulation is typically 30-40g of high-quality protein, depending on body mass and protein source. Beyond this, additional protein at that meal contributes to oxidation and other metabolic functions rather than further increasing synthesis rate.
Leucine: The Anabolic Trigger
Leucine is the primary branched-chain amino acid responsible for activating the mTOR signalling pathway that initiates muscle protein synthesis. The threshold dose is approximately 2-3g per serving. Plant proteins generally have lower leucine content per gram than animal proteins, which is why plant protein sources often require higher total doses to achieve equivalent anabolic stimulus.
Whey protein has the highest leucine content of commercial protein sources and the fastest absorption kinetics, making it particularly effective post-exercise. Casein, by contrast, is slower-digesting and better suited to extended periods without protein intake, such as overnight.
Timing Still Matters
The post-exercise period remains important. Muscle protein synthesis is elevated for 24-48 hours after resistance training, but the rate of synthesis in the immediate post-exercise window is acutely highest. Consuming a leucine-rich protein source within the first two hours post-exercise is a well-supported strategy regardless of whether you prioritise total daily intake.
The Practical Protocol
Distribute protein across 4-5 meals. Aim for 30-40g of high-quality protein per serving. Ensure at least 2.5-3g leucine per serving. Prioritise post-exercise timing. And account for protein source quality, not just grams.
The total still matters. But the details are where adaptation actually happens.
